Ponceau S is a red, water-soluble, anionic diazo dye that is made by coupling diazotized
4-ami,1’-azobenzene-3,4’-disulfonic acid (Acid Yellow 9) with 2-naphthol-3,6-disulfonic acid(R acid)
and converting the coupled product to the tetrasodium salt.
Ponceau S is a negative stain, which binds to the positively charged amino groups of the protein and is
widely used in gel electrophoresis. It also binds non-covalently to non-polar regions in the protein.
Ponceau S may not be as sensitive of a protein stain as Coomassie Blue. It is superior to Acid Fuchsin in
the VanGieson stain.
Preparation Instructions :
It is soluble in water (1 mg/ml), yielding a clear solution.
Procedure:
A solution as low as 1% ponceau S can be made in 7% acetic acid (w/w) for staining polyacrylamide gels.
The gel can be left in this solution for eight hours. However, prior to placing the gel in this staining mixture,
the proteins on the gels should be fixed with a fixative agent such as methanol:acetic acid or TCA.
Destaining of the gel can occur in 7% (v/v) acetic acid:water mixture with many changes of the
destaining solution.
Storage/Stability:
Recommended storage is at room temperature. The shelf-life should be at least two years.
